From the Pike, the Auburn exit is #10, and 290 (close!) goes right into
Worcester.
Route 2 would be the longer (time-wise) way to go.

I don't know about the best way, but the easiest way is to hop
across the river into Vermont, take I-91 south to the Mass. Pike and the
Pike east to Auburn. At the Auburn exit head north (I490 or 690; can't
remember the right number) and it takes you right into downtown Worcester (5
miles or so).
It might be shorter to exit I91 at Route 2 in Mass. and head east but it's a
slower trip and Route 2 is loaded with speed traps.
